Isolation of extraintestinal pathogenic Escherichia coli from diarrheic dogs and their antimicrobial resistance profile BJM

From January to December 2006, 92 Escherichia coli isolates from 25 diarrheic dogs were analyzed by screening for the presence of adhesin-encoding genes (pap, sfa, afa), hemolysin and aerobactin genes. Virulence gene frequencies detected in those isolates were: 12% pap, 1% sfa, 10% hemolysin and 6.5% aerobactin. Ten isolates were characterized as extraintestinal pathogenic E. coli (ExPEC) strains; all showed a multidrug resistance phenotype that may represent a reason for concern due the risk of dissemination of antimicrobial resistant genes to the microbiota of human beings.

Isolation of Shiga toxigenic Escherichia coli from butcheries in Taquaritinga city, State of São Paulo, Brazil BJM

Shiga toxigenic Escherichia coli (STEC) has been implicated as the cause of several human diseases. Samples (ground beef, grinding-machines and human hands) from 23 butcheries were assayed for E. coli using standard microbiological methods, and 287 isolates were submitted to polymerase chain reaction for the detection of stx 1, stx 2 and eae genes. Four STEC isolates were recovered, two from ground beef and two from grinding-machines; all harbored the stx 2 gene and were negative for the eae gene. All E. coli isolates including the four STEC were screened for antibiotic resistance. Les études sanitaires et la pêche à pied professionnelle : l'exemple de Bonne Anse en Charente-Maritime ArchiMer

Tout coquillage mis en vente est issu d’une zone de production (gisement naturel ou concession) classée et surveillée au niveau sanitaire. Quatre classements sont définis de A à D par ordre décroissant de salubrité, du classement A (bonne qualité) autorisant la commercialisation directe des coquillages, au classement D (très mauvaise qualité). Pour définir ce classement, une étude sanitaire est préalablement conduite par l’Ifremer sur demande de l’Administration. L’étude sanitaire réalisée à Bonne Anse en Charente-Maritime, a abouti en 2009 à un arrêté préfectoral classant en B le gisement de palourdes. Aujourd’hui, la qualité sanitaire de la zone est surveillée régulièrement par l’Ifremer, et environ 25 pêcheurs à pied professionnels exploitent le gisement.

Atomic Force Microscopy (AFM) can be used to obtain high-resolution topographical images of bacteria revealing surface details and cell integrity. During scanning however, the interactions between the AFM probe and the membrane results in distortion of the images. Such distortions or artifacts are the result of geometrical effects related to bacterial cell height, specimen curvature and the AFM probe geometry. The most common artifact in imaging is surface broadening, what can lead to errors in bacterial sizing. Nitrogen and phosphorus present in sewage can be used for microalgae growth, possibiliting cost reduction in the production of microalgae at the same time that it decreases the eutrophication potential of the effluent. This research aimed at monitoring the native community of microalgae and coliform bacteria in a secondary effluent from anaerobic municipal sewage treatment. Two treatments (aerated and non-aerated) were performed to grow microalgae under semi-controlled conditions in semi-closed photobioreactors in a greenhouse. Microbiologia de queijo tipo Minas Frescal produzido artesanalmente Ciência Rural

Com o objetivo de avaliar as condições higiênico-sanitárias do queijo Minas Frescal produzido artesanalmente em Cuiabá - MT, foi realizada análise microbiológica de trinta amostras obtidas em dois pontos de comercialização. A análise microbiológica consistiu da contagem em placas de Staphylococcus aureus e da contagem estimativa de coliformes totais, fecais e Escherichia coli pela técnica do NMP-3 tubos. Na determinação de coliformes fecais, 28 amostras (93,33%) apresentaram número mais provável (NMP) > 10² NMP/g e somente duas amostras (6,67%) estavam dentro dos padrões legais exigidos. Na contagem de S. aureus, em 29 amostras (96,67%) obteve-se valores superiores a 10³ufc/g, estando apenas 1 amostra (3,33%) em conformidade com o padrão legal....

Modeling the pressure inactivation dynamics of Escherichia coli BJMBR

Escherichia coli, as a model microorganism, was treated in phosphate-buffered saline under high hydrostatic pressure between 100 and 300 MPa, and the inactivation dynamics was investigated from the viewpoint of predictive microbiology. Inactivation data were curve fitted by typical predictive models: logistic, Gompertz and Weibull functions. Weibull function described the inactivation curve the best. Two parameters of Weibull function were calculated for each holding pressure and their dependence on holding pressure was obtained by interpolation. With the interpolated parameters, inactivation curves were simulated and compared with the experimental data sets.

Motilidade espermática e integridade acrossomal em doses de sêmen suíno refrigeradas e inoculadas com Esecherichia coli e Staphylococcus aureus Ciência Rural

Foram utilizados 12 ejaculados, coletados de maneira asséptica, distribuídos em sete tratamentos, sendo um grupo controle e os demais inoculados com três diferentes concentrações de S. aureus ou E. coli (5 x 10(5), 5 x 10(6) e 5 x 10(7) UFC/ml). Durante 96 horas, foram avaliados a motilidade espermática, o percentual de acrossomas intactos (NAR), o pH e o número de unidades formadoras de colônia (UFC/ml) das bactérias inoculadas. O desenvolvimento bacteriano foi decrescente ao longo das 96 horas de armazenamento. Multidrug-resistant urinary tract isolates of escherichia coli from Ribeirão Preto, São Paulo, Brazil BJID

Multiple resistances to antimicrobial drugs arising in Escherichia coli isolates may complicate therapeutic management of urinary tract infection (UTI) by this organism. In order to assess the multidrug resistance (MDR) among urinary E. coli isolates, we have tested 11 antimicrobial drugs against 67 isolates from outpatients attended in a tertiary-care teaching hospital and of 78 isolates from a municipal health unit, respectively in Ribeirão Preto, State of São Paulo, Brazil. The antimicrobial sensibility of Escherichia coli strains isolated from the uterine content of bitches was evaluated. Fifteen E. coli strains were tested in relation to their susceptibility to different antimicrobials. The results demonstrated 100% of resistance to all tested drugs, being a quite conflicting finding compared to other works, which observed variable resistance of those bacteria to different antimicrobials but not the same multi-resistance pattern. The detection of those multi-resistance strains configures a problem, with important implications on the antimicrobial therapy. Therefore, additional investigations for a best characterization and extension of this problem are needed.

New vectors derives from pUC 18 for clonig and thermal-induced expression in Escherichia coli BJM

We report the construction of two vectors for Escherichia coli: pUC72, for molecular cloning, and pPLT7, for thermal-induced expression. The main feature of pUC72 is a novel polylinker region that includes restriction sites for Nde I and Nco I which provide an ATG codon for proper translation initiation of expressed genes. Vector pPLT7 is ideal for thermo-inducible expression in host cells that carry the cI857 repressor gene. The use of pPLT7 was validated by the successful expression of the genes encoding carp and porcine growth hormones. These vectors provide novel cloning possibilities in addition to simple, non-expensive, high level expression of recombinant proteins in E. coli.
